For.Airborne particles of polymeric carbon black (PCB) and black carbon (BC) are ubiquitous in urban environments. With recent research on the role of aerosols in respiratory health, it is of great importance to study the health effects of these particles. In this study, we investigated the inflammatory response elicited by airborne particles of black carbon (BC) and carbon black (CB) of black and non-black origin, as well as their ability to induce leukocyte adhesion to the pulmonary endothelium in vivo. Furthermore, we studied the phagocytic ability of neutrophils by monitoring their ingestion of particles that are not recognized by the respiratory defense system, and we measured bacterial uptake by phagocytes in vivo. BC induced a concentration-dependent increase in IL-8 release in a human lung epithelial cell line in vitro. CB also caused a significant but lower dose-dependent increase in IL-8 production. The deposition and inflammatory response to particles of black and non-black origin were strongly correlated. BC from black origin displayed a significantly enhanced inflammatory response in comparison with particles of black or non-black origin. CB, regardless of the origin, elicited an inflammatory response comparable to that of BC. We also found that both polymers increased leukocyte adhesion to the pulmonary endothelium in vivo. Interestingly, BC from black origin elicited an increased leukocyte adhesion in vivo, while CB from both black and non-black origin induced a comparable and significantly lower adhesion. We found a positive correlation between the amount of particles that adhered to the pulmonary endothelium, the number of phagocytosing leukocytes, and the surface area of particle deposition. Importantly, BC was more efficient than CB in promoting bacterial uptake, which was significantly enhanced only when BC was of black origin. These findings strongly suggest that the origin of airborne particles of black carbon is important for their inflammatory response in vitro and in vivo. Furthermore, we found that uptake by phagocytes was strongly dependent on the deposition and inflammatory response elicited by particles, since particles that were more efficiently phagocytosed also 3da54e8ca3
